Plant DNA isolation: Twice the DNA yield in less time

Molecular studies of plants often depend on high-quantity and high-quality DNA extractions. This can be quite difficult in plants, however, due to a diversity of compounds and physical properties found in plants. "Tannins, tough fibrous material, and/or secondary compounds can interfere with DNA isolation," explains Dr. Thomas Givnish, principal investigator of a new study published by Jackson Moeller et al. in the October issue of Applications in Plant Sciences. This is further complicated by the fact that different plant groups have different compounds and, therefore, different DNA extraction methods may be necessary to obtain sufficient high-molecular-weight DNA depending on the species you are working with. Moeller, Givnish, and their colleagues at the University of Wisconsin-Madison evaluated a new DNA isolation technique and compared it to two often-used DNA extraction methods for plant tissues. The new technique is based on MagnaCel paramagnetic cellulose particles (PMC), and was originally designed and developed for forensic applications in which only minute amounts of DNA are present. The utility of the PMC method was compared with both a silica column technique (DNeasy Plant Mini Kit) and the cetyltrimethylammonium bromide (CTAB) method. DNA helps police solve six breaking and entering cases

Carl Preston Crawford Jr. GREENSBORO, N.C. A series of commercial breaking and entering cases from May has been solved after evidence collected from a scene was matched to DNA processed by a contracted forensics laboratory. Carl Preston Crawford Jr., 27, of 1800 Longfellow St., is charged with breaking and entering into six SUBWAY restaurants throughout Greensboro after evidence collected from one of the crime scenes matched a known sample of DNA processed through Cellmark Forensics BioTracks DNA database. Crawford previously was arrested on Aug. 20 and charged with breaking and entering into nearly a dozen Radio Shack stores across North Carolina during August and September. Four of those stores were in Greensboro. After Cellmark Forensics, a business contracted by the City of Greensboro to do DNA testing, notified the Greensboro Police Department that the SUBWAY evidence matched Crawfords DNA, detectives took out additional warrants for his arrest on Sept. 29. In May 2014, the Greensboro Police Department entered into a two-year contract with Cellmark Forensics for the analysis, search, match, and retrieval of local DNA samples. DNA ‘bias’ may keep some diseases in circulation, biologists show

It's an early lesson in genetics: we get half our DNA from Mom, half from Dad. But that straightforward explanation does not account for a process that sometimes occurs when cells divide. Called gene conversion, the copy of a gene from Mom can replace the one from Dad, or vice versa, making the two copies identical. In a new study published in the American Journal of Human Genetics, University of Pennsylvania researchers Joseph Lachance and Sarah A. Tishkoff investigated this process in the context of the evolution of human populations. They found that a bias toward certain types of DNA sequences during gene conversion may be an important factor in why certain heritable diseases persist in populations around the world. Lachance is a postdoctoral fellow at Penn in Tishkoff's lab and will be starting his own lab at Georgia Tech in January. Tishkoff is a Penn Integrates Knowledge Professor with appointments in the Perelman School of Medicine's Department of Genetics and the School of Arts & Sciences' Department of Biology.
