A study with mice suggests the drugs could act like a bad electrician, causing neurons to be miswired and interfering with their ability to send messages to the brain. The findings, from the scientist whose original research led to the drug development, are published in the journal Molecular Neurodegeneration and will be presented Saturday, Feb. 18, at the 2012 annual meeting for the American Association for the Advancement of Science in Vancouver. "Let's proceed with caution," said Robert Vassar, professor of cell and molecular biology at Northwestern University Feinberg School of Medicine. "We have to keep our eyes open for potential side effects of these drugs." Ironically, he says, the drugs could impair memory. The drugs are designed to inhibit BACE1, the enzyme Vassar originally discovered that promotes the development of the clumps of plaque that are a hallmark of Alzheimer's. The Food and Drug Administration (FDA) is reviewing Qnexa again, after turning down its submission for approval two years ago ("FDA Turns Down Weight Loss Drug Qnexa, But Makers See Short Timeline For Approval"). There is an FDA Advisory Panel (Endocrinologic and Metabolic Drugs Advisory Committee) meeting on 22 February. The Panel will look at 2-years' worth of clinical data. When Qnexa was last reviewed, the Agency only had 1-year's worth of data to look at. The Advisory Panel consists of experts, none of whom work for the FDA. Martin, MD Feb. 16, 2012 -- An experimental, implanted drug-delivery microchip that releases medication on command from an external wireless control could one day free patients from daily injections and improve treatment compliance. Results from the first human study of the programmable microchip were reported Thursday in Vancouver at the annual meeting of the American Association for the Advancement of Science -- 15 years after researchers at Massachusetts Institute of Technology (MIT) first came up with the idea for the device. If future research is promising, the technology could be used to treat a wide range of conditions that require frequent or daily injections, says Robert Farra, the study's author and chief operating officer of the company developing the drug-delivery device, MicroCHIPS Inc. FDA raises safety concerns for Qnexa diet pill

By Dow Jones February 17, 2012 WASHINGTON -- The Food and Drug Administration raised concerns about possible long-term side effects of Vivus Inc.'s diet pill Qnexa in a review posted on its website Friday. The documents posted in advance of an advisory panel meeting on Wednesday, cited concerns about potential side effects of the drug such as birth defects and an unknown impact on the heart, but said Qnexa was effective at helping people lose weight. The pill is being reviewed by the FDA for a second time after previously being rejected by the agency in 2010. The review of the drug includes two-year clinical data rather than one year's worth of data that was reviewed in 2010. Qnexa will face the agency's endocrinologic and metabolic drugs advisory committee, which is made up of non-FDA medical experts. The panel is being asked to vote on and discuss a series of questions including whether the "overall benefit-risk assessment" supports approval of Qnexa. The U.S. Food and Drug Administration has ignited rampant discussion about the possibility, after announcing its plan to review the treatment known as Qnexa. The reason for all the excitement? Clinical trials have shown that people who take Qnexa lose significant amounts of weight; a trial published in 2009 showed patients on the drug lost an average of nearly 17 kilograms (37 pounds). More related to this story The FDA rejected approval of the drug in 2010, however, because of nagging concerns it may increase the risk of birth defects and heart problems. On Friday, the FDA reiterated its stance that Qnexa may cause serious side effects. Cancer drug may reverse effects of Alzheimer's

SALT LAKE CITY &#8212 Researchers have discovered that a drug used to treat cancer may have benefits for sufferers of Alzheimer's disease. According to neuroscientists at Case Western Reserve University, use of the drug Bexarotene in mice led to quick reversals of pathological, cognitive and memory deficits caused by the disease. Bexarotene has been Food and Drug Administration-approved for the treatment of cancer for more than a decade. Now, researchers say there is good reason to look into approving it for the treatment of Alzheimer's disease. When treated with the drug, soluble amyloid levels in mice fell by 25 percent within six hours. The effects lasted for up to three days. Soluble amyloid buildup in the brain is what causes the memory impairments that are symptoms of Alzheimer's, scientists believe. Also within three days of being administered with the drug, the mice showed marked behavioral improvement. Researchers found that their nesting instinct, which seemed to have disappeared in Alzheimer's-diseased mice, reappeared in mice treated with the drug. By Brian Orelli | More Articles February 17, 2012 | The headlines make the briefing documents for the Food and Drug Administration advisory panel reviewing VIVUS' (Nasdaq: VVUS  ) Qnexa sound horrible: "FDA raises safety concerns for Vivus diet pill" "Vivus obesity drug has safety risks -- US FDA staff" But the 3% increase the market awarded VIVUS today more accurately reflects the information in the briefing documents for the panel next week: No new issues is good news. The FDA is still concerned with the oral clefs seen in babies of women taking Johnson & Johnson's epilepsy medication Topamax, one of the active ingredients in Qnexa. VIVUS doesn't need the advisory panel to say that there isn't a risk; I doubt it will, considering the data. The panel of experts just needs to conclude that the risk is low enough that educating doctors not to prescribe it to pregnant patients will solve the problem. The FDA has already lightened up on the issue, requesting a change from excluding women who are capable of getting pregnant to just those that are currently or are planning to become pregnant.
