Thermo Fisher Scientific Targets Neurons and Primary Cells with Expansion of Transfection Reagent Portfolio
Posted: Published on November 4th, 2014
CARLSBAD, Calif.--(BUSINESS WIRE)--Researchers can now more efficiently study predictive cell models and human disease with the launch of a new high-efficiency transfection reagent for mRNA delivery in neurons and primary cell types. Lipofectamine MessengerMAX offers up to a five times the efficiency of DNA reagents and enables researchers to easily transfect biologically relevant cell models without electroporation or viruses. Researchers studying these cells have been faced with challenges due to limited options in the market. DNA reagents tend to be inefficient for transfection of primary cells, and other approaches can introduce toxicity into cells and negatively affect data. Primary cell models such as neurons, hepatocytes, keratinocytes and fibroblasts provide researchers with more meaningful answers to their research questions. To transfect these cells, Lipofectamine MessengerMAX uses novel lipid nanoparticle technology optimized to deliver the highest amount of mRNA possible without the need to enter the nucleus. "In my lab, we used Lipofectamine MessengerMAXto successfully transfect mesenchymal stem cells with high efficiency without the toxic effects of viral vectors, said Daniel Cohen, Ph.D., Research Specialist at the University of Pennsylvania. The ability to easily transfect primary cells and circumvent the difficulties of transgenic DNA delivery that can impede discovery enables us to … Continue reading

Decoding the emergence of metastatic cancer stem cells
Posted: Published on November 1st, 2014
From our community In the first study of its kind, Rice University researchers have mapped how information flows through the genetic circuits that cause cancer cells to become metastatic. The research reveals a common pattern in the decision-making that allows cancer cells to both migrate and form new tumors. Researchers say the commonality may open the door to new drugs that interfere with the genetic switches that cancer must flip to form both cancer stem cells and circulating tumor cells -- two of the main players in cancer metastasis. "Cells have genetic circuits that are used to switch certain behaviors on and off," said biophysicist Eshel Ben-Jacob, a senior investigator at Rice's Center for Theoretical Biological Physics and co-author of a new study in the Journal of the Royal Society Interface. "Though some of the circuits for metastasis have been mapped, this is the first study to examine how cancer uses two of those circuits, in concert, to produce not just cancer stem cells, but also dangerous packs of hybrid stem-like-cells that travel in groups to colonize other parts of the body." Metastasis -- the spread of cancer between organs -- causes more than 90 percent of cancer deaths, but … Continue reading

Mechanism that allows differentiated cell to reactivate as a stem cell revealed
Posted: Published on October 31st, 2014
One kind of stem cell, those referred to as 'facultative', form part -- together with other cells -- of tissues and organs. There is apparently nothing that differentiates these cells from the others. However, they have a very special characteristic, namely they retain the capacity to become stem cells again. This phenomenon is something that happens in the liver, an organ that hosts cells that stimulate tissue growth, thus allowing the regeneration of the organ in the case of a transplant. Knowledge of the underlying mechanism that allows these cells to retain this capacity is a key issue in regenerative medicine. Headed by Jordi Casanova, research professor at the Instituto de Biologa Molecular de Barcelona (IBMB) of the CSIC and at IRB Barcelona, and by Xavier Franch-Marro, CSIC tenured scientist at the Instituto de Biologa Evolutiva (CSIC-UPF), a study published in the journal Cell Reports reveals a mechanism that could explain this capacity. Working with larval tracheal cells of Drosophila melanogaster, these authors report that the key feature of these cells is that they have not entered the endocycle, a modified cell cycle through which a cell reproduces its genome several times without dividing. "The function of endocycle in living … Continue reading
