Abstract Introduction Stem cell therapy has recently been introduced to treat patients with type 2 diabetes mellitus (T2DM). However, no data are available on the efficacy and safety of allogeneic Whartons Jelly-derived mesenchymal stem cell (WJ-MSC) transplantation in patients with T2DM. Here we performed a non-placebo controlled prospective phase I/II study to determine efficacy and safety of WJ-MSC transplantation in T2DM. Twenty-two patients with T2DM were enrolled and received WJ-MSC transplantation through one intravenous injection and one intrapancreatic endovascular injection (catheterization). They were followed up for 12months after transplantation. The primary endpoints were changes in the levels of glycated hemoglobin and C-peptide and the secondary endpoints included insulin dosage, fasting blood glucose (FBG), post-meal blood glucose (PBG), inflammatory markers and T lymphocyte counts. WJ-MSC transplantation significantly decreased the levels of glucose and glycated hemoglobin, improved C-peptide levels and beta cell function, and reduced markers of systemic inflammation and T lymphocyte counts. No major WJ-MSC transplantation-related adverse events occurred, but data suggest a temporary decrease in levels of C-peptide and beta cell function at one month after treatment, possibly related to intrapancreatic endovascular injection. There Are Cells Inside Teeth That Can Turn Back Into Stem Cells

Inside each of our hard, calcified teeth is a small population of living stem cells that can differentiate into many types of tissue. The origin of those stem cells has long been unknown, but scientists may now have a completely surprising answer: Cells of the nervous system can migrate into the middle of a tooth and actually turn back into stem cells. If verified, this could be a possible new source for stem cells. Teeth are connected to nerves, as anyone who's had the misfortune of a toothache would know. Igor Adameyko of the Karolinska Institute in Stockholm and his team were studying glial cells, which support and protect neurons, in mice. By fluorescently labeling these glial cells, they could track the fate of these cells over time. What they found was entirely surprising. Glial cells from nerves in the gums actually migrated into teeth, where they turned into mesenchymal stem cells and eventually into tooth cells. Stem cells differentiating into specific cells is to be expected, but glial cells turning into stem cells is the exact opposite of what we thought we knew. Dr. Defeng Zou and co-workers from the First Affiliated Hospital of China Medical University, China focuses on the effect of miRNA overexpression on the differentiation of bone marrow-derived mesenchymal stem cells into neurons. In the study released on the Neural Regeneration Research (Vol. 9, No. 12, 2014), researchers used GeneChip technology to analyze the expression of miRNAs in bone marrow-derived mesenchymal stem cells, neural stem cells and neurons. They constructed a lentiviral vector overexpressing miR-124 and transfected it into bone marrow-derived mesenchymal stem cells. Intracellular expression levels of the neuronal early markers -III tubulin and microtubule-associated protein-2 were significantly increased, and apoptosis was reduced in transfected cells. After miR-124-transfected bone marrow-derived mesenchymal stem cells were transplanted into the injured rat spinal cord, a large number of cells positive for the neuronal marker neurofilament-200 were observed in the transplanted region. The Basso-Beattie-Bresnahan locomotion scores showed that the motor function of the hind limb of rats with spinal cord injury was substantially improved. Photo: Facebook Supporters of a Brisbane mother-of-two who died while undergoing a controversial stem cell treatment in Russia say it did not cause her death, nor have others been discouraged from seeking it. Kellie van Meurs suffered from a rare neurological disorder called stiff person syndrome, which causes progressive rigidity of the body and chronic pain. She travelled to Moscow in late June to undergo an autologous haematopoietic stem cell transplant (HSCT) under the care of Dr Denis Fedorenko from the National Pirogov Medical Surgical Centre. Scientists injected stem cells prior to encouraging mice to perform exercises considered the rough equivalent of human lengthening contractions performed as part of resistance training. They found the time of muscle repair was shortened and they achieved considerable development of the mices muscles. We have an interest in understanding how muscle responds to exercise, and which cellular components contribute to the increase in repair and growth with exercise, says study author and University of Illinois kinesiology and community health professor Marni Boppart. Boppart says the research is important in treating atrophy and age-related degradation in both the structure and the function of muscles.
