There are currently over 300 clinical trials evaluating MSC therapeutic utility in a variety of diseases. Unlike other types of cellular therapies, MSCs can be used in allogeneic settings without immunosuppressive therapy due to the cells ability to evade immune detection. MSCs home to injured tissue and provide therapeutic support through a multifaceted mechanism. They secrete a dynamic assortment of bioactive cytokines, trophic factors, and anti-inflammatory molecules in response to environmental cues. The traditional sources of MSCs are from adult tissues and have limited expansion capacity and so must be constantly replenished from more donors, and screened for pathogens. Moreover, there is an appreciable loss of potency upon propagation of adult MSCs in culture, which along with inconsistent quality of current MSC sources, limits the scalability of their use in therapy. We believe our new method can overcome these limitations as we have taken advantage of a versatile precursor cell called the hemangioblast. hESC-derived hemangioblasts replace the need for donors, and our pre-clinical studies have demonstrated these hESC-MSCs have the capacity to respond to environmental cues, influence immune cell function, and exert therapeutic effects to reduce clinical symptoms in two different autoimmune disease models. GDNF transfection promotes neuronal differentiation of bone marrow mesenchymal stem cells

PUBLIC RELEASE DATE: 21-Mar-2014 Contact: Meng Zhao eic@nrren.org 86-138-049-98773 Neural Regeneration Research Studies have shown that the differentiation rate of grafted bone marrow mesenchymal stem cells into mature neuron-like cells is very low. Therefore, it is very important to establish an effcient and stable induction protocol to promote the differentiation of bone marrow mesenchymal stem cells into neuron-like cells in vitro and elucidate the mechanisms underlying differentiation for the treatment of central nervous system diseases. Jie Du and colleagues from Sichuan University in China found that glial cell line-derived neurotrophic factor/bone marrow mesenchymal stem cells have a higher rate of induction into neuron-like cells, and this enhanced differentiation into neuron-like cells may be associated with up-regulated expression of glial cell line-derived neurotrophic factor, nerve growth factor and growth-associated protein-43. The researchers provide experimental support for the therapeutic use of glial cell line-derived neurotrophic factor gene-modified bone marrow mesenchymal stem cells in transplantation strategies for central nervous system diseases. The Philippines is the biggest market for the popular, if highly controversial, alternative treatment in Germany called fresh-cell therapy (FCT). Fresh cells derived from the fetus of an unborn lamb are injected into patients, and are said to cure a large number of illnesses. Despite the high cost of the treatment, wealthy Filipinos are undeterred, and typically arrive in droves in a sleepy town outside Frankfurt, their hopes of being cured or rejuvenated pinned on the life of every donor sheep. Given its renown, its no surprise that questions about the efficacy and safety of FCT has been the subject of discussions among health professionals. There have also been rumors of deaths after FCT. The proponents of FCT in Germany, however, claim that all talk about patient deaths and questionable safety standards are unfounded, and an uncouth effort to discredit FCT so that the same medical professionals here could promote stem-cell therapy, which is allowed in the country. Lawrence Jyh Liew added an answer: How can I eliminate contamination in my human MSCs culture? I believe they are mainly yeast infections due to the white film that develops in the wells but even when that is not present I seem to get high cell death when culturing human MSCs on thermanox with PRP. Will alterations in LINC proteins change response? Lab studies show how MSC respond to the dynamic physical environment. We are trying establish a protocol in order to obtain MSC from cord tissue according protocols described in different papers. Although most of bibliography described that is easy procedure, we are having many difficulties. We cant obtain cells using explants method. We already did 17 cord tissue and we cant see any cells. What can be we are doing wrong? I created a new protocol to derive MSC from human induced pluripotent stem cells. I characterized the cells using anti-CD73 and anti-CD105. I got 80 to 90% positive cells for the two markers CD73/CD105. I wanted to know if this two markers are sufficient to conclude that my cells are MSC? This is an interesting question! Traumatic brain injuries (TBI), sustained by close to 2 million Americans annually, including military personnel, are debilitating and devastating for patients and their families. Regardless of severity, those with TBI can suffer a range of motor, behavioral, intellectual and cognitive disabilities over the short or long term. Sadly, clinical treatments for TBI are few and largely ineffective. In an effort to find an effective therapy, neuroscientists at the Center of Excellence for Aging and Brain Repair, Department of Neurosurgery in the USF Health Morsani College of Medicine, University of South Florida, have conducted several preclinical studies aimed at finding combination therapies to improve TBI outcomes. In their study of several different therapies -- alone and in combination -- applied to laboratory rats modeled with TBI, USF researchers found that a combination of human umbilical cord blood cells (hUBCs) and granulocyte colony stimulating factor (G-CSF), a growth factor, was more therapeutic than either administered alone, or each with saline, or saline alone. Among them are chairman of the Philippine Medical Association Leo Olarte, PSSCM treasurer and legal counsel; Bu Castro, secretary; Rey Melchor Santos, president; Oscar Tinio, vice president; and Jose Asa Sabili, chairman. In a statement, PRC Chairperson Teresita Manzala on Sunday said she directed the Professional Regulatory Board of Medicine (PRBOM) to initiate, investigate, and file charges against the five doctors before the PRCs legal division for unprofessional, dishonorable, and unethical conduct. Earlier on 10 January 2014, the SEC cancelled the registration of the PSSCM for submitting a fabricated document. In an order signed by SEC Acting Director Ferdinand Sales, the commission said the PSSCM had committed fraud in procuring its Certificate of Incorporation for its application for corporate registration.
