The ultimate source of these cells is called the hematopoetic stem cell or HSC for short. One feature of an HSC is self- renewal, the ability to divide and still remain a stem cell. HSCs can also differentiate into more mature cells and this transformation is driven by cytokines (protein messengers) and growth factors. The best known source for HSCs is bone marrow. Along with HSCs in the marrow are other stem cells, the mesenchymal stem cells or MSCs. Multiple experiments have shown that HSCs are very plastic, meaning they can be coaxed into helping with the regeneration and repair of non-blood tissue. This has led to the idea that "adult" HSCs and MSCs, if placed in the proper environment can serve as a "repair shop" for connective tissues such as cartilage, muscle, and tendon, ligament, and bone. Washington, January 31 (ANI): The Idu-Mishmi community of Arunachal Pradesh and RIWATCH (Research Institute of World's Ancient Traditions Cultures and Heritage) have made a great contribution in studying dormant Mycobacterium in TB that has affected nearly 4 billion people in the world and causing 1.9 million deaths yearly. TB is a huge problem in India. The incidence of this infectious disease is much more higher in Northeast of India and especially Arunachal Pradesh than rest of India. The doctors prescribe drug treatment for months together to the TB patients, however TB recurs leading to death and suffering. It was not clear as to how the bacteria of TB hide in the body from months of drug treatment. New Delhi, Jan. 31: Scientists, using bone marrow samples from members of the Idu Mishmi tribe in Arunachal Pradesh, have discovered the hideout in the human body where tuberculosis bacteria can lie dormant for years. A collaborative study by Indian and US scientists has identified stem cells in the bone marrow that seem to serve as a "protective niche" for TB germs to evade the drugs' action and the human immune system. The study, published yesterday in the journal Science Translational Medicine, relied on samples of bone marrow provided by volunteers among Idu Mishmi tribals who live in a remote mountain range in Arunachal's Lower Dibang Valley. By volunteering to participate in the study, scientists say, this indigenous community has contributed to efforts to resolve a long-standing medicine mystery. While effective antibiotics have been available to treat and cure TB for more than 50 years, TB bacilli remain in the body despite a successful course of treatment and can cause the disease to recur after years or even decades. Tuberculosis can 'lurk' in patients' bone marrow

Traces of the bacteria were identified in mesenchymal stem cells, which can produce specialised cell types including bone, fat and cartilage. Although the stem cells are typically found in the bone marrow they are also capable of moving to the lungs the ideal environment for tuberculosis bacteria. After testing their theory on mice in a previous study, the researchers carried out a small clinical trial in India where bone marrow biopsies were taken from nine patients who had been treated for tuberculosis and who had no traces of the bacteria in their respiratory tract. In eight of the nine patients, bacterial DNA could still be found in bone marrow stem cells, and in two of these cases living bacteria were found. Dr Bikul Das, lead author of the study, said: "Not only is this strong evidence that the tuberculosis can remain dormant in stem cells, but it shows that the living bacteria could be recovered from these cells after a long period of time. Tuberculosis Hiding in Bone Marrow Helps Deadly Bacteria Survive

Tuberculosis can hide in peoples bone marrow, lying dormant after aggressive drug therapy and emerging years later to re-infect, according to a finding that may help explain why the disease is difficult to eradicate. One third of the worlds population, more than 2.2 billion people, are infected with the deadly bacteria that causes TB. A four-drug cocktail of antibiotics and other drugs are typically prescribed for a minimum of six months in an effort to wipe out the disease. Still, most remain vulnerable even after treatment. Investigators writing today in Science Translational Medicine said they have uncovered the first evidence of tuberculosis nestled in mesenchymal stem cells in the bone marrow of people treated for the disease. The bacterias hideout in the self-renewing cells, where they capitalize on protection from the bodys own immune system, may explain how the germs survive. The next step is to find out how a re-infection is triggered, and then how to stop it, researchers said.
