Phase III trial will investigate potential of statins as treatment for multiple sclerosis – The Pharmaceutical Journal

The potential of the cheap cholesterol-lowering drug simvastatin as a treatment option for multiple sclerosis is to be investigated in a phase III trial involving more than 1,000 patients. The trial will be led by Jeremy Chataway, a researcher at the UCL Institute of Neurology, who conducted a previous smaller phase II trial, the results of which were published in The Lancet[1] in 2014 and suggested that simvastatin appeared to be a treatment option worthy of further investigation. The phase III trial will involve 1180 patients with secondary progressive MS, for which there are currently no licensed treatments that can slow or stop disability progression, who will be recruited by 30 centres across the UK. The trial will take six years to complete, with the first patients receiving treatment later this year. Chataway said: This drug holds incredible promise for the thousands of people living with secondary progressive MS in the UK, and around the world, who currently have few options for treatments that have an effect on disability. "I woke up one morning and all of my toes were numb and tingling, and it felt like I had rocks in my shoes," described Arterbridge. She had multiple sclerosis. "I have a lesion on my spine, and I have three on my brain," Arterbridge detailed. Dr. Michael Racke, a neurologist at The Ohio State University's Wexner Medical Center in Columbus, Ohio, is an expert in MS and one of a nationwide team of researchers studying the effects of ocrelizumab. "Almost half the patients had no evidence of disease activity in terms of their MS," Racke said. "That's much higher than we've seen with any other MS treatment." Ocrelizumab is also the first drug ever available for the primary, progressive form of the disease, which includes about 15 percent of MS patients. Statinscould soon be prescribed to treat multiple sclerosis after a groundbreaking trial began to see if the cheap cholesterol busting drug could delay symptoms. The 6 million projectis being led by University College London and involves 1,180 people at 30 centres across Britain. It follows a successful small trial in 2014 which showed a significant reduction in the rate of brain shrinkage and an increase in mobility after two years for people on high doses of simvastatin , which usually prescribed to lower cholesterol. Charities said the trial was a 'momentous step' forward which offered hope to thousands of people with relapsing MS. The trial is scheduled to last six years, but because statins are already known to be safe and widely used, the treatment would quickly pass through regulators if it was deemed to be successful. The FDA has approved a new drug to treat people living with multiple sclerosis. The chronic auto-immune disorder causes a patients own system to attack his or her brain and spinal cord. The drug is not a cure for MS, but the treatment will allow some patients to live virtually symptom-free for the first time in years.
