Study of live human neurons reveals Parkinson's origins

ScienceDaily (Feb. 7, 2012) — Parkinson's disease researchers at the University at Buffalo have discovered how mutations in the parkin gene cause the disease, which afflicts at least 500,000 Americans and for which there is no cure. The results are published in the current issue of Nature Communications. The UB findings reveal potential new drug targets for the disease as well as a screening platform for discovering new treatments that might mimic the protective functions of parkin. UB has applied for patent protection on the screening platform. "This is the first time that human dopamine neurons have ever been generated from Parkinson's disease patients with parkin mutations," says Jian Feng, PhD, professor of physiology and biophysics in the UB School of Medicine and Biomedical Sciences and the study's lead author. As the first study of human neurons affected by parkin, the UB research overcomes a major roadblock in research on Parkinson's disease and on neurological diseases in general. The problem has been that human neurons live in a complex network in the brain and thus are off-limits to invasive studies, Feng explains. Laser targeting could help fight Parkinson's

SAN FRANCICSO (KGO) -- Innovative research at San Francisco's Gladstone Institutes could lead to a new treatment for fighting Parkinson's disease. Research using lasers is pointing the way to a new target for drugs. Dr. Anatol Kreitzer has spent years shedding new light on the neural pathways of Parkinson's disease. In fact, when ABC7 first met him, he was using lasers to coax the neurons into firing, allowing him to map them. "The first step was to identify which circuits are actually affected in Parkinson's disease, and that's what we did," he said. That laser mapping allowed him to move to the next logical step, looking for a way to interrupt the malfunctioning circuits that cause the symptoms of Parkinson's. After crunching data on hundreds of candidates, his team at San Francisco's Gladstone Institutes zeroed in on a tiny protein called RGS4. It regulates signaling in the part of the brain responsible for movement. "RGS4 becomes overactive in Parkinson's disease and it actually causes these circuits to become overactive as well," Kreitzer explained. But the team was also looking for a cause and effect. NPH diagnosis in San Tan Valley man returns him to normal life

Surgery to implant a shunt into a 75-year-old San Tan Valley man's brain has returned him to a nearly normal life years after he was diagnosed with Parkinson's disease and began suffering from bladder incontinence and dementia.  Edwin "Ed" Stall had been diagnosed with Parkinson's disease-like symptoms in 2007 at the age of 71. It was hard for him to walk and his balance was off. An MRI showed "nothing alarming," Stall said. "I always felt a sensation like I was falling forward when I was walking," he said. After falling a few times, the doctor tried to correct Edwin's gait, or his stance, as he walked. "He's always walked faster than me, because he's taller," his wife of 54 years said. "Then I started walking faster than he did." Over the next three years, Stall and his wife, Betty continued to seek answers to his medical problems to no avail. Slowly, he stopped being able to do work around the house or play a round of golf, he said. He was unable to walk down the street to a friend's house and instead had to have Betty drive the car. Researchers visualize the development of Parkinson's cells

New technology from the University of Bonn in Germany lets researchers observe the development of the brain cells responsible for the disease. Up until now, research into the brain cells responsible for Parkinson's disease has focused on the function and degeneration of these neurons in the adult and aging brain. The new tissue slicing method, which will be published in the world's only peer-reviewed science video journal, the Journal of Visualized Experiments (JoVE), allows scientists to observe the development of these brain cells for the first time. "Little is known about the behavior of these neurons during their differentiation and migration phase," said article author Dr. Sandra Blaess, "and with this technique, we can really observe how these cells behave during development."
