Human primordial cells created in the lab

A cell programming technique developed at the Weizmann Institute turns them into the earliest precursors of sperm and ova IMAGE:These are clusters of human embryonic stem cells that were differentiated to an early germ cell (PGC) state (colored cells). Each color reveals the expression of a different gene. (l-r)... view more Credit: Weizmann Institute of Science Groups at the Weizmann Institute of Science and Cambridge University have jointly managed the feat of turning back the clock on human cells to create primordial germ cells - the embryonic cells that give rise to sperm and ova - in the lab. This is the first time that human cells have been programmed into this early developmental stage. The results of their study, which were published today in Cell, could help provide answers as to the causes of fertility problems, yield insight into the earliest stages of embryonic development and potentially, in the future, enable the development of new kinds of reproductive technology. "Researchers have been attempting to create human primordial germ cells (PGCs) in the petri dish for years," says Dr. Jacob Hanna of the Institute's Molecular Genetics Department, who led the study together with research student Leehee Weinberger. PGCs arise within … Continue reading

'Tissue chips' could replace animal studies, UW-Madison researchers say

Posted: Published on December 21st, 2014

Drugs that seem safe in animal studies are sometimes found to be harmful in humans. The health effects of many chemicals on the market in products such as cosmetics, food additives and industrial and household cleaners arent fully known. Theres a huge need to come up with better ways of testing compounds to see if theyre toxic, said James Thomson, the UW-Madison stem cell pioneer. Thomson, who first grew human embryonic stem cells in a lab in 1998 and co-discovered a way to reprogram mature cells to their embryonic state in 2007, is working with researchers across campus on a new quest. Theyre creating tissue chips clusters of interacting cells that mimic specific organs, such as a model of a developing brain. Using stem cells, miniature scaffolds and sophisticated computer programs, theyre crafting prototypes that could someday replace animal testing for drugs and serve as screening tools for environmental toxins. About 80 percent of experimental drugs fail in human clinical trials because they are unsafe or ineffective, with some 30 percent found to be toxic in people despite promising results in animal studies, according to the National Institutes of Health. Largely because of the expensive failures, it costs more than … Continue reading

Stem Cell Scandal Scientist Haruko Obokata Resigns

Posted: Published on December 20th, 2014

A Japanese Stem Cell Scientist At The Heart Of A Scandal Over False Claims And Fabricated Research Has Resigned. Dr Haruko Obokata published supposedly groundbreaking research showing stem cells could be made quickly and cheaply. There were irregularities in data, no other group in the world could repeat her findings and her own university concluded it could not be done. In a statement Dr Obokata said: "I even can't find the words for an apology." Stem cells can become any other type of tissue and hold great potential in medicine. They are already being investigated to heal the damage caused by a heart attack and to restore sight. But they are expensive and difficult to produce and one source - embryos - raises serious ethical questions. 'Major discovery' Dr Obokata's scientific paper published in the prestigious journal Nature claimed that stem cells could be produced from normal adult cells by dipping them into acid for a 30-minute shock period. The announcement of the creation of these "Stap" cells (stimulus-triggered acquisition of pluripotency) sent shockwaves around the world.
