Stem cell behavior of human bowel discovered for first time

For the first time, scientists have uncovered new information on how stem cells in the human bowel behave, revealing vital clues about the earliest stages in bowel cancer development and how we may begin to prevent it. The study, led by Queen May University of London (QMUL) and published today in the journal Cell Reports, discovered how many stem cells exist within the human bowel and how they behave and evolve over time. It was revealed that within a healthy bowel, stem cells are in constant competition with each other for survival and only a certain number of stem cells can exist within one area at a time (referred to as the 'stem cell niche'). However, when investigating stem cells in early tumours, the researchers saw increased numbers of stem cells within each area as well as intensified competition for survival, suggesting a link between stem cell activity and bowel cancer development. The study involved studying stem cells directly within the human body using a specially developed 'toolkit'. The toolkit worked by measuring random mutations that naturally accrue in aging stem cells. Single-cell analysis holds promise for stem cell and cancer research

UC San Francisco researchers have identified cells' unique features within the developing human brain, using the latest technologies for analyzing gene activity in individual cells, and have demonstrated that large-scale cell surveys can be done much more efficiently and cheaply than was previously thought possible. "We have identified novel molecular features in diverse cell types using a new strategy of analyzing hundreds of cells individually," said Arnold Kriegstein, MD, PhD, director of the Eli and Edythe Broad Center of Regeneration Medicine and Stem Cell Research at UCSF. "We expect to use this approach to help us better understand how the complexity of the human cortex arises from cells that are spun off through cell division from stem cells in the germinal region of the brain." The research team used technology focused on a "microfluidic" device in which individual cells are captured and flow into nano-scale chambers, where they efficiently and accurately undergo the chemical reactions needed for DNA sequencing. The research showed that the number of reading steps needed to identify and spell out unique sequences and to successfully identify cell types is 100 times fewer than had previously been assumed. Discredited Stem Cell Scientist Found Hanged

TOKYO, Japan - An eminent biologist linked to a discredited stem cell research project was found hanged at his workplace, police said Tuesday. The body of Yoshiki Sasai was discovered by security guards at the Riken Center for Developmental Biology in Kobe City, Japan, shortly before 9 a.m. local time (10 p.m. Monday ET). Several letters addressed to his colleagues were found tucked in his briefcase, broadcaster NHK reported. "It's very, very unfortunate," Riken's deputy director Shigekazu Saito told reporters. Sasai, 52, supervised a team of scientists, led by Haruko Obokata, that announced in the journal Nature in January that they had found a game-changing way to make stem cells using ordinary mouse cells. However, suspicions were raised over Obokatas research and she was accused of falsifying data. In June, Obokata agreed to retract her findings. Although the Riken Centers investigation cleared Sasai of wrongdoing, he was reprimanded for his failure to properly supervise Obokata's work. In April, he apologized for allowing an incident that might taint Japans reputation for scientific research. Japan scientist in discredited stem cell research commits suicide

Tokyo: A Japanese researcher at the centre of discredited research that was initially hailed as a potential breakthrough for stem cell treatment, killed himself after months of stress and exhaustion, officials said on Tuesday. Yoshiki Sasai, co-author of the high-profile research that had seemed to offer hope for replacing damaged cells or even growing new human organs, was found early on Tuesday at the Riken institute where he worked in Kobe, western Japan, police and the institute said. It is confirmed as a suicide, said a police spokesman. It was a hanging. Sasai, 52, had been hospitalised in March for stress and become less receptive to media inquiries during the controversy over the teams research, said Riken spokesman Satoru Kagaya. The scientist had seemed completely exhausted in their last phone conversation around May or June, Kagaya told a televised news conference. As deputy director of Rikens Centre for Developmental Biology, Sasai supervised the work of lead author Haruko Obokata, which took the world of molecular biology by storm when it was published in the British journal Nature in January. Scientist in Research Scandal Found Dead

A senior Japanese scientist embroiled in a stem-cell research scandal died Tuesday in an apparent suicide, police said. Yoshiki Sasai, who supervised and co-authored stem-cell research papers that had to be retracted due to falsified contents, was found suffering from cardiac arrest at the government-affiliated science institute RIKEN in Kobe, in western Japan, according to Hyogo prefectural police. Sasai, 52, was deputy chief of RIKEN's Center for Developmental Biology. A security guard found him with a rope around his neck, according to RIKEN. Sasai was rushed to a hospital, but was pronounced dead two hours later. Police and RIKEN said Sasai left what appeared to be suicide notes, but refused to disclose their contents. RIKEN spokesman Satoru Kagaya told a news conference that Sasai had three letters with him, each addressed to Haruko Obokata, a co-author of the research papers, as well as senior members of RIKEN and fellow researchers. Two other notes addressed to RIKEN officials were on Sasai's secretary's desk. Sasai's health had deteriorated over the past few months, and he had been receiving medical treatment.
