Murdered mother-of-three Kylie Blackwood.

Homicide investigators have taken DNA samples from criminals living in a Melbourne suburb on the day Kylie Blackwood was killed.

August 1 marks one year since the mother-of-three was killed in her McCaffrey Rise home in Pakenham in what police believe was a "burglary gone wrong".

Homicide Squad Detective Senior Sergeant Stuart Bailey told Fairfax Media investigators are reducing their pool of suspects through the "elimination DNA sampling" of hundreds of persons of interest.

The list of people police want to take DNA samples from include burglars and drug users who were living in Pakenham on the day Mrs Blackwood was killed.

Detective Senior Sergeant Bailey said a mysterious man wearing a hooded, striped jumper seen both before and after Mrs Blackwood was killed and the use of her bankcard after her death pointed to a "burglary gone wrong".

Mrs Blackwood left the garage door open when she returned home that morning and a man in a horizontal-striped hoodie, dark sunglasses and a baseball cap was seen walking away from the house 20 minutes later.

The same man was seen peering into the McCaffrey Rise house before Mrs Blackwood arrived home.
